A complete virus-mediated system for recombinant AAV productionincluding: (a) a first recombinant adenovirus encoding one or more AAVREP78/68 polypeptides and one or more viral capsid polypeptides; (b) asecond recombinant adenovirus encoding a gene of interest and includingAAV inverted terminal repeats, wherein the AAV inverted terminal repeatsflank the gene of interest; (c) viral helper functions; and (d) a host celltransduced with the first recombinant adenovirus, the second recombinantadenovirus, and the viral helper functions.
