PURPOSE: Provided is a DNA cassette which has base sequences encoding a biologically active peptide and a method which produces a bio peptide using microorganism transformed by an expression vector having DNA cassette CONSTITUTION: A DNA cassette which has base sequences encoding multimer of bio peptide and linker of peptide cut by protease or chemical resolve agent is provided by ligation reaction of more than two basic units, DNA(ideally between 2 and 10). An expression vector is obtained by a cloning method that is inserting the DNA cassette into a vector and the DNA cassette vector transforms a microorganism. The transformed microorganism is cultivated and the DNA cassette produces a multimer of peptide. The multimer peptide from culture solution, is cut by protease and chemical resolve agent and produces a biologically active peptide or a modification of bio peptide
