PROBLEM TO BE SOLVED: To provide an improved method for stainning a slide, using immunochemical reagent capable of minimizing preparation of the agent and applicable for improving a chemical method for immune tissue manually and automatically and applicable to its many improvement for associated immunochemical e.g. enzyme-linked immunosorbent assay(ELISA) and immunofluorescence assay and hybridization in-situ.;SOLUTION: This technique comprises washing an assay region (region including tissue slice) of a slide with water and a detergent, covering the assay region by applying thereto an evaporation suppressing liquid, combining the tissue slice of an antigen requiring unmasking with a solution of an improved and stabilized protease, rinsing the slide, reapplying the evaporation suppressing liquid, combining a primary antibody in an improved diluent containing globulin from the same kind of a second antibody with the tissue fragment for a sufficient time practically completing combining antibodies, rinsing the slide and reapplying the evaporation suppressing liquid.
