Manufacturing method of a nucleic acid fragment (DNA or RNA) labeled 3¿, comprising the enzymatic incorporation of nucleotide derivative having as precursor a compound of formula: wherein R1 represents a nucleic base and R2 represents a group corresponding to one of the following formulas: - (CH2) n-NH2 - (CH2) n-SH - (CH2) n-COOH - (CH2) n-OH - (CH2) n-NH-R3 - (CH2 ) n-SR3 - (CH2) n -CO-R3 - (CH2) n-OR3 where n is an integer ranging from 1 to 12 and R3 is a group derived from a label, a protein, a enzyme, a fatty acid or a peptide at the 3¿ OH nucleic acid fragment.
