Microfluidic devices (200, 300, 400), platforms (100), modules (500, 600) and methods are provided for rapid microscale processing in preparation of nucleic acid samples, including cell lysis, sedimentation, capture, and purification wherein the nucleic acids are analyzed with the objective of detecting and identifying unknown bioagents in environmental, clinical or other samples. Detection and identification of unknown bioagents which include bacteria, viruses and the like, is accomplished by a combination of nucleic acid amplification and molecular weight determination using primers which hybridize to conserved sequence regions of nucleic acids derived from a bioagent and which bracket variable sequence regions that uniquely identify the bioagent.
