By using the sampling system, a controller determines the position of a sampled surface and controls a translation positioning mechanism (300) to adjust the position of a clamp (200) until the first end surface of a sampling head (100) is in contact with the sampled surface; an extractant injection mechanism (400) injects an extractant into a first flow channel (104); a droplet transferring and transporting mechanism (500) injects air into a second flow channel (105) at an interval of a first preset time, and sample droplets are led out by a third flow channel (106); a translation positioning mechanism (300) drives a fixture (200) to move at an interval of a second preset time so that the sampling head (100) is separated from the sampled surface. Since the sampling head (100) is in direct contact with the sampled surface, automatic sampling can be implemented without separation, thereby improving sampling efficiency. In addition, the use of droplet extraction will not cause damage to the sampled surface, and a sampling cavity (103) is close to the sampled surface, so as to increase the concentration of sample droplets and reduce the possibility of loss of key compound information, thereby improving sampling quality.
