PURPOSE: Full-length genomic RNA of Japanese encephalitis virus, infectious JEV cDNA therefrom, and the use thereof are provided. The JEV cDNA is useful for identifying JEV gene and studying molecular biological mechanism related to replication, transcription and translation of JEV. CONSTITUTION: Full-length genomic RNA of Korean type Japanese encephalitis virus comprises 95 bp of 5'-untranslated region, 10,299 bp of polypeptide coding region, and 574 bp of 3'-untranslated region, has the nucleotide sequence set forth in SEQ ID NO: 15, and contains 5'-terminal nucleotide sequence of 1AGAAGT- and 3'-terminal nucleotide sequence of -GATCT10968. Infectious JEV cDNA from the full-length genomic RNA of JEV comprises a promoter, SP6 or T7, upstream to the 5'-terminal of JEV genomic RNA, and a restriction enzyme recognition site, XhoI or XbaI, downstream to the 3'-terminal of JEV genomic RNA. A vector pBACT7/JVFLx/XbaI(KCTC 10346BP) contains T7 promoter and JEV cDNA. A vector pBACSP6/JVFLx/XbaI(KCTC 10347BP) contains SP6 promoter and JEV cDNA.
