PURPOSE: A method for enhancing Agrobacterium-mediated genetic transformation by pre-treatment of sugar or salts is provided, thereby obtaining high efficiency of transgenic plants, so that it can be applied to transformation-hard plants. CONSTITUTION: The method for enhancing Agrobacterium-mediated genetic transformation by pre-treatment of sugar or salts comprises the steps of: inducing embryogenic callus by culturing zygotic embryo of ginseng and Cortex Acanthopanacis; preparing Agrobacterium and plasmid for genetic transformation; dipping embryogenic callus and cotyledon in 0.1M to 2M sucrose solution for 1 to 12 hours or dipping embryogenic callus and cotyledon in 0.01M to 0.2M of magnesium sulfate for 1 to 60 minutes; co-culturing the pretreated embryogenic callus and cotyledon with Agrobacterium and selecting transformed embryo using antibiotics such as kanamycin; and analyzing the gene insertion and expression by using Gus reaction.
