Isolated polynucleotide (I) that encodes a polypeptide (II) involved in resistance to Aphis gossypii and/or viral transmission by this aphid, also the complement of (I) and sequences that hybridize selectively under stringent conditions to (I) or its complement are new. (II) is at least 80, preferably at least 95, % identical with a 1473 amino acid sequence (3), reproduced. Independent claims are also included for: (1) a fragment (Ia) containing at least 10 bp of (I), its complement or hybridizing sequences; (2) polynucleotide (Ib), encoding a polypeptide with same properties as (II), that has been obtained by screening a genomic DNA or cDNA library from a plant, using a probe or primer pair derived from (I) or (Ia); (3) genetic marker (GM) for detecting presence/absence of an allele of the Vat (= virus aphid transmission resistance) gene that favors resistance to A. gossypii and viral transmission by this aphid, which is any of markers L273, L246, V681, V1684, V432, GRP805, M8, marker D and marker E; (4) nine primer pairs (all sequences reproduced) for amplification of GM; (5) kit for detecting, in plants, the specified Vat allele that includes at least one primer pair of (4); (6) method for detecting resistance/susceptibility of a plant to A. gossypii and viral transmission by this aphid, by detecting the specified Vat allele; (7) expression cassette containing (I) or (Ia) and a promoter; (8) recombinant vectors that contain (I), (Ia) or the cassette of (7); (9) cells genetically transformed by (I) or (Ia); (10) transgenic plants transformed by (I); and (11) (II) as a new compound.
