A silenced anti-CD28 antibody defective in mitogenic activity which has a mutation or modification in the Fc receptor binding site of the anti CD28 antibody is described. The antibody can be used in the manufacture of a medicament for inducing T-cell tolerance, immunosuppression or for treating organ or tissue transplant rejection. Methods of producing the antibody, polynucleotides encoding the antibody, vectors and isolated host cells are also described.
