PURPOSE: A composition for screening antibiotics comprising a gene spr0603 from Streptococcus pneumoniae or a protein encoded thereby, and a method for screening antibiotics using the same are provided. The gene and protein encoded thereby are essential to survival of Streptococcus pneumoniae, and the antibiotics screening method against Streptococcus pneumoniae is useful for screening and developing antibiotics against other pathogenic bacteria. CONSTITUTION: The composition for screening antibiotics comprises a gene spr0603 from Streptococcus pneumoniae or a protein encoded therein, wherein the gene spr0603 has the nucleotide sequence of SEQ ID NO:1, or the nucleotide sequence of SEQ ID NO: 1 wherein one or more mutations selected from disruption, deletion, insertion, point, substitution, nonsense, missense, polymorphism and rearrangement mutation are occurred; and the protein encoded by the gene spr0603 is UDP-N-acetylmuramoyl-L-alanine-D-glutamate ligase having the amino acid sequence of SEQ ID NO:2 or a polypeptide fragment expressed from the nucleotide sequence of SEQ ID NO: 1 wherein one or more mutations selected from disruption, deletion, insertion, point, substitution, nonsense, missense, polymorphism and rearrangement mutation are occurred. The method for screening antibiotics comprises contacting the composition with a testing material, and determining whether the expression of gene spr0603 is inhibited.
