A aminoglycoside phosphotransferase, a gene encoding the same enzyme and uses thereof are provided. An aminoglycoside phosphotransferase gene transfers tolerance against aminoglycoside antibiotics to a microorganism producing aminoglycoside antibiotics and activates phosphorylation enzymes, so that the gene is useful for mass-producing antibiotics from a recombinant microorganism containing the aminoglycoside phosphotransferase gene. The gene encoding the aminoglycoside phosphotransferase, isolated from Micromonospora purpurea(ATCC 15835) has the nucleotide sequence set forth in SEQ ID NO:1. The aminoglycoside phosphotransferase encoded by the gene of SEQ ID NO:1 has the amino acid sequence set forth in SEQ ID NO:2. A recombinant expression vector comprises the aminoglycoside phosphotransferase gene of SEQ ID NO:1. A transformant producing the aminoglycoside phosphotransferase is produced by transforming a microorganism producing aminoglycoside antibiotics with the recombinant expression vector. A cosmid comprises the aminoglycoside phosphotransferase gene of SEQ ID NO:1. A method for producing antibiotics comprises culturing the transformant producing aminoglycoside antibiotics and having tolerance against aminoglycoside antibiotics in a medium.
