Generation of Large Genomic DNA Deletions

The method of the present invention uses gene targeting to delete large fragments of genomic DNA using substitutional targeting constructs. This substitutional targeting construct may comprise a selectable marker and is constructed to include two sequence regions homologous to the 5 'and 3' flanking sequences of the targeted locus. After the targeting construct is transfected into the desired cell line, the gene target-mediated deletion is selected for identification and further characterization. Examples of suitable loci are immunoglobulin genes including MHC class I and II antigens and variable or constant regions of kappa, lambda or heavy chains.
