CO-EXPRESSION VECTOR OF HSV-TK AND PNP ENZYMES, PREPARATION METHOD THEREOF AND USE OF THE VECTOR IN GENE THERAPY

PURPOSE: A co-expression vector of HSV-TK(herpes simplex virus thymidine kinase) and PNP(purine nucleoside phosphorylase) enzymes, a preparation method thereof and the use thereof in gene therapy are provided. A transformed cell with the co-expression vector when the transformed cell is treated with GCV has more improved cell-death effect than each HSV-TK and PNP enzymes. CONSTITUTION: A vector HSV-TK-ires-PNP(KCTC 10205BP) co-expressing HSV-TK and PNP enzymes comprises (a) a promoter suitable for a target cell, (b) a DNA sequence encoding HSV-TK protein, (c) an ires site, (d) a DNA sequence encoding PNP protein, and (e) a poly-A signal, wherein the promoter suitable for a target cell is CMV(cytomegarovirus) promoter. A method for preparing the vector HSV-TK-ires-PNP(KCTC 10205BP) co-expressing HSV-TK and PNP enzymes comprises the steps of: (a) PCR amplifying a plasmid containing the HSV-TK gene using a primer; (b) PCR amplifying a plasmid containing the PNP gene using a primer; and (c) cloning the PNP gene into a pIRES vector, and cloning the HSV-TK gene.
