Procedure random recombination in vitro nucleic acid fragments, comprising the following steps: a) fragmentation of a library of polynucleotide sequences b) hybridization of the fragments obtained on an assembling template c) ligation of the fragments hybridized having adjacent ends by means of a ligase d) denaturation of the ligated fragments of the assembling template, and repetition of the hybridization steps (b), ligation (c) and denaturation (d) to form several random recombinants, said process being further characterized in that steps (a) to (d) are performed in the absence of polymerase.
