A protein chip for screening competitors of microphthalmia-associated transcription factor(MITF) and a screening method of MITF competitors by using the same chip are provided to screen compounds inhibiting expression of melanin formation associated enzymes by inhibiting the binding between a promoter of melanin formation associated enzymes and MITF, so that target molecules related to skin whitening are developed. The protein chip for screening competitors of microphthalmia-associated transcription factor(MITF) comprises a substrate coated with beta-cyclodextrin(beta-CD) and MBP(maltose-binding protein)-MITF hybrid. A kit for screening the competitors of microphthalmia-associated transcription factor(MITF) comprises the protein chip and a MITF-specific DNA, wherein the MITF-specific DNA is labeled by a fluorescent material selected from FITC(fluorescein isothiocyanate), TRITC(tetramethylrhodamine isothiocyanate), YFP(yellow fluorescent protein), RFP(red fluorescent protein), DAPI(diamidine-2-phenylindole), Cy2, Cy3 and Cy5; and the MITF-specific DNA is E-box(CATGTG). The screening method of MITF comprises the steps of: (1) adding a candidate material into the protein chip; and (2) analyzing the binding inhibition between the MITF-specific DNA and MITF.
