PPROBLEM TO BE SOLVED: To provide a method for developing protein vaccines and vaccines for avian influenza using a reverse genetic method. PSOLUTION: The present invention relates to a preparation method of protein vaccines, and comprises the steps of: (a) providing at least one amino acid sequence of an epitope peptide of a target antigen protein; (b) modifying a wild-type nucleic acid sequence of the epitope peptide; (c) synthesizing a plurality of primers of the modified nucleic acid sequence; (d) synthesizing the modified nucleic acid in vitro; (e) binding the modified nucleic acid to form a plasmid; (f) transforming the plasmid into a host cell to produce a synthesized nucleic acid encoded with the epitope peptide; and (g) collecting and purifying the epitope peptide.
