MODEL ANIMAL FOR DIABETIC RETINOPATHY AND NEPHROPATHY

PPROBLEM TO BE SOLVED: To obtain a model mammal developing diabetic complication, which is useful for elucidating mechanism of pathogenesis of diabetes caused in the case of occurrence of continuous activation of calcium/calmodulin-dependent protein kinase 2 and for screening a therapeutic agent for diabetic complication. PSOLUTION: A transgene in which two genes of a gene encoding calcium/calmodulin-dependent protein kinase 2 having 1 amino acid substitution mutation and a poly A addition-SV40 gene are bonded to the downstream of rat insulin gene promoter is constructed. The transgene is microinjected into the male pronucleus of a mouse fertilized egg, the obtained egg cells are cultured and transplanted into the uterine tube of pseudopregnant female mouse. Then the transplanted animal is bred, a body mouse having the transgene is selected as a transgenic mouse from the born baby mice and the transgenic mouse is bred for several mouths to develop diabetic complication.
