A tyrosine phenol-lyase variant is provided to have increased activity and thermostability compared to a wild-type tyrosine phenol-lyase, thereby being useful as a biological catalyst and useful for industrially producing tyrosine. The thermostable tyrosine phenol-lyase variant has an amino acid sequence described as SEQ ID : NO. 6. The method for preparing the variant comprises the steps of: (a) culturing a transformed microorganism obtained by introducing an expression vector including a gene encoding the thermostable tyrosine phenol-lyase variant into a host cell such as bacteria, fungus and yeast so as to express the thermostable tyrosine phenol-lyase variant; and (b) isolating the thermostable tyrosine phenol-lyase variant from the thermostable tyrosine phenol-lyase variant expressed microorganism.
