Recombinant Coxsackievirus Vector and Replication Deficient Coxsackieviruses with No Cytotoxicity

A recombinant coxsackievirus vector and replication deficient coxsackieviruses with no cytotoxicity are provided to deliver a foreign gene specifically to myocardial cells without side effects on the myocardial cells. A recombinant coxsackievirus vector contains the genomic nucleotide sequence of coxsackievirus in which VPO, VP3 and VP1 nucleotide sequences are deleted, and at least one gene of coxsackievirus selected from 2A, 2B, 2C, 3A, VPg, 3C and 3D, or further contains 5'-NTR(non-translating region) and a nucleotide sequence encoding an amino acid sequence of SEQ ID NO:2, located at downstream of the 5'-NTR, wherein a foreign gene which is located at downstream of the nucleotide sequence encoding the amino acid sequence of SEQ ID NO:2 is further inserted into the deleted VPO, VP3 and VP1 nucleotide sequences. The replication deficient coxsackieviruses with no cytotoxicity contains the genomic nucleotide sequence of coxsackievirus in which VPO, VP3 and VP1 nucleotide sequences are deleted and is manufactured by (a) preparing the recombinant coxsackievirus vector containing the genomic nucleotide sequence of coxsackievirus in which VPO, VP3 and VP1 nucleotide sequences are deleted, (b) preparing a vector containing the VPO, VP3 and VP1 nucleotide sequences, and (c) co-transfecting a cell with the vectors of steps(a) and (b) to obtain the replication deficient coxsackieviruses with no cytotoxicity which is encapsidated with the VPO, VP3 and VP1 proteins.
