PPROBLEM TO BE SOLVED: To provide a method for detecting the mismatch of a double-stranded nucleic acid in improved efficiency and accuracy. PSOLUTION: A method determines the presence of a variation in a nucleic acid sequence by using a protein binding a single-stranded DNA, the above method wherein the presence of the variation in the nucleic acid sequence is determined by using a nucleic acid amplification reaction product to use the protein binding a single-stranded DNA, and a kit for determining the presence of the variation in the nucleic acid sequence.
