ITGA1(integrin A1) gene polymorphisms associated with bone mineral density(BMD) and fracture risk are provided to predict susceptibility to osteoporosis or fracture risk of a subject, so that the gene polymorphisms are useful as marker of osteoporosis genetic predisposition. An ITGA1 gene polymorphism marker for predicting susceptibility to osteoporosis is +73187CT(exon 3), +76969TG(intron 5), or their haplotype BL1_ht1, wherein +73187th base is C and +76969th base is T or BL1_ht2, wherein +73187th base is T and +76969th base is G which is associated with BMD of femoral neck and femoral shaft; +159174AC(intron 28) or its BL3_ht1 , wherein +159174th base is C, +164104th base is C and +165697th base is T which is associated with fracture risk; or BL2-ht3, wherein +109100th base is T, +110140th base is G, and +130991st base is C which is associated with BMD of Ward's triangle. A method for predicting susceptibility to osteoporosis comprises the steps of: (1) collecting a DNA sample from a subject; (2) detecting the ITGA1 gene polymorphism marker from the DNA sample; and (3) identifying the detected ITGA1 gene polymorphism.
