METHOD FOR DISCRIMINATING KIND OF CYNODON DACTYLON WITH MICROSATELLITE MARKER

PROBLEM TO BE SOLVED: To provide a method for discriminating the kind of Cynodon dactylon by the use of a genotype which is a genotype at a specific microsatellite site in the genome DNA of control Cynodon dactylon and the genotype specific to the kind of the control Cynodon dactylon, as an indicator, and the like.;SOLUTION: Microsatellites are extracted from ESTs originated from the aboveground portion of Cynodon dactylon, and a primer is designed so that 90 to 300 bp PCR fragments containing the microsatellites can be obtained. Various kinds of Cynodon dactylon have been subjected to PCRs using a genome DNA as a template and using the designed primer pair. The PCR products have been subjected to fluorescent label reactions and then separated by an electrophoresis method using a 10% polyacrylamide gel. Consequently, it has found that a kind-specific PCR fragment length can be detected and used as an indicator to determine the kind of a Cynodon dactylon to be tested.;COPYRIGHT: (C)2010,JPO&INPIT
