PROBLEM TO BE SOLVED: To provide a method for detecting the variation (mt3243 variation) in the 3243 site of mitochondrial DNA.;SOLUTION: A nucleic acid probe has a region containing mt3243 variation amplified by PCR (polymerase chain reaction), is labeled its terminal with a fluorescent dye and decreases the fluorescence intensity of the fluorescent dye by hybridization. The variation is detected by using the nucleic acid probe having a base sequence of 14-40 base length and starting from the base number 230 of a base sequence expressed by the sequence number 2 and labeled its 3'-terminal with the fluorescent dye, determining the fluorescence of the fluorescent dye to perform the melting curve analysis and detecting the variation based on the result in the melting curve analysis.;COPYRIGHT: (C)2005,JPO&NCIPI
