The present invention relates to a process for the preparation of acetic acid esterase from ragi malt. Ragi (Eleusine coracana L.), apart from being economically cheaper and abundantly available cereal, is an important source of acetic acid esterase. In the present invention, detergent (Triton X-100), phenolic adsorbent (polyvinylpolypyrrolidine), reducing agent (reduced glutathione) and defined concentrations of metal ions (Ca2+and Mg2+) are used to yield high acetic acid esterase activity. In this invention a-naphthyl acetate is used as the substrate to monitor the acetic acid esterase activity which is quantified spectrophotometrically at 235nm for the release of a-Naphthol from a-Naphthyl acetate. Acetic acid esterase obtained from this process exhibits the activity in the range of 0.044 to 10.21 micro moles per minute.
