The present invention relates to a method of genotyping skipjack tuna by analysis of polymorphisms in the number of microsatellite DNA repeats. The repeat sequence is amplified by the use of specific primers. The number of repeats, and therefore the distance between the primers, is highly variable in a population, thereby providing an allelic marker for the locus. The combined information from multiple loci provides a means of distinguishing individuals for parentage/paternity testing, forensic testing, inbreeding, larval, juvenile and adult identification, individual relatedness, population structure analysis, quantitative trait loci analysis, mapping genes of interest such as genes controlling diseases that are genetically linked to the marker loci and in toxicology and ecotoxicological studies. Accordingly, the invention relates to the PCR procedure using an effective amount of nucleotide, subserving PCR reagents, fluorescently labeled primers with and without M13 tails, and amplification parameters that have been identified according to the invention. Five pairs of highly informative novel microsatellite primers for amplification of tunas microsatellite loci particularly skipjack tuna Katsuwonus pelamis are disclosed.
