Fast and biocompatible method of extraction, saponification and concentration of polyunsaturated fatty acids (PUFAS) due to differences in biomass solubility. The invention provides a method for preparing a concentrate of polyunsaturated fatty acids (PUFAS) from biomass comprising: (a) extracting and saponifying the biomass by treatment with a lower alcohol and an alkali metal hydroxide at 75-90 ° C; (b) cooling the mixture obtained in (a) to obtain a liquid phase and a solid phase; (c) filter the solid phase at the cooling temperature; (d) acidify the filtrate; (e) extracting the acidified filtrate with an organic solvent; and (f) purify the extract by distilling the solvent; wherein in step (a) the lower alcohol is absolute ethanol and the alkali metal hydroxide is solid sodium hydroxide; and the cooling of stage (b) and the filtration of stage (c) are carried out at a temperature of 0-4 ° C. The PUFAS concentrate thus obtained can be used in the agri-food, pharmaceutical or chemical industry.
