The invention is located in the field of coagulation diagnosis and relates toan in vitro method for determining vonWillebrand factor (VWF) activity in a sample. The method includes the use of amutant gain-of-function variant of the GPIb.alpha. protein,thus making it possible to dispense with the use of ristocetin, botrocetin oranother ristocetin- or botrocetin-equivalent substance. Theinvention further relates to a method for determining von Willebrand factor(VWF)-cleaving activity of the ADAMTS-13 protease.
