Detecting an analyte in a biological sample comprises using a complex of the type A-B-C-D, where A is a solid phase, B and C are components of a reversible binding pair and D is an analyte binder. The method comprises adding the sample to the complex so that the analyte is bound to D, removing the sample, adding a release buffer to break the bond between B and C, and detecting the analyte in the release buffer. Independent claims are also included for: (1) device for carrying out the above assay, comprising an A-B-C-D complex as above; (2) kit for carrying out the above assay, comprising an A-B complex and a C-D complex and a release buffer.
