PROBLEM TO BE SOLVED: To provide a measuring method for quantifying and standardizing cancer antigen-specific IgG (Immunoglobulin G) antibody titer existing in various cancer patients' sera.;SOLUTION: In this standardization method, capturing side antibody, rather than the IgG of an analyte, is serially-diluted, highly-concentrated (in saturated amount) human IgG is used, at a stage of analyte addition to saturate the capturing antibody with the human IgG so that effects from cross-reactivity with detecting side antibody and IgG mixing may be ignored. First, reaction of solid-phased anti-human antibody with purified human IgG is used, instead of antigen in order to mimic complex of solid-phased antigen and antibody in serum. In this case, an excess amount of purified human IgG is added to the prepared and solid-phased concentration series of anti-human antibody, instead of adding concentration series of the purified human IgG to the solid-phased anti-human antibody, generating the saturated complex. This operation is to avoid rise of background caused by cross-reactivity between enzyme label anti-human antibody to be used later and the solid-phased anti-human antibody.;COPYRIGHT: (C)2011,JPO&INPIT
