METHOD FOR DETERMINATION OF ACTIVITY OF MATRIX METALLOPROTEINASE-2

A method for determination of activity of matrix metalloproteinase-2 (MMP-2) consists in that the MMP-2 is extracted from the tissue, than the activity of MMP-2, which is contained in tissue supernatant, is determined by the hydrolysis of protein substrate at the temperature 37 ° in a definite time. The products of hydrolysis are determined by means of colorimetric method. The tissue supernatant, containing MMP- the volume of 0.1-0.2 ml is incubated with 0.4-0.8 ml of gelatin solution in NaCl water bath during 30-45 minutes without preliminary activation with aminophenmercuriacetate. The reaction products are determined by spectrophotometer at the wave length of 570 nm.
