A method of in situ detecting an immunohistochemical epitope or nucleic acid sequence of interest in a biological sample, comprising binding a labeled conjugate molecule with an enzyme to the epitope or sequence of interest in the presence of a reductive species redox activity and ion soluble metal, wherein said reductive species redox activity is a substrate for said labeled conjugate molecule with an enzyme and wherein said enzyme of said labeled conjugate molecule catalyzes the transformation of inactive reductive species redox a reducing agent, thereby reducing the metal to insoluble metal particles in oxidation state 0, at or about the point where the enzyme is anchored ion.
