摘要
PURPOSE: A gene structure containing gene encoding two or more reprogramming-inducing factors is provided to simultaneously express plural reprogramming-inducing factors and obtain induced pluripotent stem cells(iPS cells) with high efficiency.;CONSTITUTION: A gene structure contains genes encoding two or more reprogramming-inducing factors. The genes are mediated with DNA segment encoding a peptide of sequence number 1. The gene structure contains a gene encoding Oct3/4, gene encoding Nanog, and gene encoding Sox2. The genes are mediated with DNA segment encoding the peptide of sequence number 1. The gene structure contains a sequence of sequence number 15. A plasmid which expresses multiple reprogramming-inducing gene comprises the gene structure. The plasmid is obtained by recombining the gene structure to viral vector plasmid. A method for preparing induced pluripotent stem cells(iPS cells) comprises: a step of introducing the gene structure to human-derived somatic cells of immortalized cell line; a step of culturing the somatic cells or immortalized cell line; and a step of isolating embryonic stem cell-like colonies from culture.;COPYRIGHT KIPO 2010
