A COLORIMETRIC ASSAY FOR SULFIREDOXIN ACTIVITY USING INORGANIC PHOSPHATE DETECTION

PURPOSE: A method for measuring sulfiredoxin activity is provided to quantitatively measure activity of sulfiredoxin and to enable massive activity measurement. CONSTITUTION: An organic phosphate released by ATP hydrolysis is measured by measuring inorganic phosphate during reduction of hyperoxidized peroxiredoxin. A method or measuring sulfiredoxin comprises: a step of preparing reagent A containing ammonium molybdate tetrahydrate and H_2SO_4; a step of dissolving polyvinyl alcohol to the distilled water and adding malathite green carbinol hydrochloride to prepare reagent B; a step of mixing a sample with the reagent A at room temperature; a step of adding the reagent B to the mixture and mixing; and a step of measuring absorbance at 630nm.
