Rapid Assays for Detection of Potential Biological Warfare Threats Using PCR Combined with Colorimetric Detection

In the previous research program (Contract no: N68171-96-C-9106) PCR/ DNA probe based assays were designed for the detection of the toxigenic clostridia and V. cholerae. The PCR assays were combined with a colorimetric membrane based detection system incorporating specific DNA probes to facilitate the rapid and sensitive detection of these pathogens. Investigation of sample preparation methods for environmental samples was undertaken with successful application of the C. perfringens PCR assay for the detection of C. perfringens in water and soil samples. The objectives of this research program included: (a) the formulation of the colorimetric membrane assays into a more standardised format, (b) the development of multi-probe screening as says for the different pathogen groups by combining genus specific PCR with a panel of species specific probes, (c) Investigation and development of preparation methods for application to environmental samples, and (d) Application of the membrane-based colorimetric assay technology in environmental samples.
