The PCR-supported method for differential detection of physiological condition of cells comprises achieving the differentiation through the selection of suitable PCR primers for amplification of a section of a specific target sequence, so that at least one of the oligonucleotide, used as PCR-primer, has an area beginning from the 3'-end extending over a large number of nucleotide bases with identical sequence or sequence homology distinct to a double stranded cDNA as to the genomic DNA containing the specific target sequence. The PCR-supported method for differential detection of physiological condition of cells comprises achieving the differentiation through the selection of suitable PCR primers for amplification of a section of a specific target sequence, so that at least one of the oligonucleotide, used as PCR-primer, has an area beginning from the 3'-end extending over a large number of nucleotide bases with identical sequence or sequence homology distinct to a double stranded cDNA, which is formed through the reverse transcription of a post-transcriptionally modified RNA of the specific target sequence, as to the genomic DNA containing the specific target sequence. The differential detection of physiological condition is the distinction between living and dead cells. The target sequence is a protein coding gene or ribosomal RNA coding gene. The post-transcriptional modification includes removing introns by splices.
