PROBLEM TO BE SOLVED: To solve a problem of a BiFC method known as a method for assaying interaction of proteins necessitating light emission by reassociation of a fluorescent protein in the case of interaction of target proteins, comprising emission of fluorescent light even in the absence of interaction when affinity of fragments of the fluorescent protein is stronger than the affinity between the target proteins.SOLUTION: An amino acid of a fragment of a fluorescent protein binding to the protein to be observed is partially substituted with other amino acid to control the affinity of the fragment of the fluorescent protein to the level of the affinity of the target proteins.
