The cascaded Raman sensing, the present invention provides a method used to analyze the content of a biological sample such as serum. Binding one or more of contacting the biological sample, a nanoporous biosensor having a probe that binds specifically to an analyte known to generate fluorescence, was coupled to the semiconductor structure of the porous I to form a complex. Fluorescence emission from the biosensor is obtained by contacting the Raman-active probe that binds specifically to the binding complex, the bound complex, it is irradiated with light in the biosensor. Emission of fluorescence of these generates a Raman signal from the bound complex. Raman signal by detecting the Raman signal generated from the binding complex is related to the protein-containing analyte bound is indicative of the presence of the protein-containing compound in the sample. The methods of the invention are useful for providing a protein profile of a patient sample. The present invention provides a detection system useful for practicing the method of the present invention.
