The present invention describes a protein with enzymatic activity of polynucleotide phosphorylase and simple and efficient methods for the purification thereof, which maintains its enzymatic activity after being purified. The polynucleotide phosphorylase may be obtained after the encoding DNA thereof is expressed in bacterial expression systems in an efficient manner and without presenting the typical problems associated with the regulation of the protein expression in bacterial systems. On the other hand, the polynucleotide phosphorylase may be easily purified by affinity chromatography using agarose NTA-Ni2 + imidazole.
