Methods and compositions for inhibition of innate immune responses and autoimmunity using immunoregulatory polynucleotides with a TGC sequence

The use of an oligonucleotide comprising less than 200 bases or base pairs and further comprising an immunosuppressive or immunoinhibitory sequence for the treating a TLR7/8-dependent immune disease. The immunosuppressive or immunoinhibitory sequence is the trinucleotide sequence TGC at the 5' end of the oligonucleotide. The oligonucleotide does not include the immunostimulatory dinucleotide sequence CG wherein the C is unmethylated. The oligonucleotide can also be used to treat TLR7/8-dependent immune response and TLR9-dependent immune response.
