A method of producing a double stranded RNA effector molecule suitable for use in inhibiting the replication of a hepatitis C virus (HCV), or an expression construct expressing said molecule, said method comprising: a) selecting the region of the non-coding strand replication intermediate corresponding to the 3 'untranslated region conserved of said HCV virus as a target; b) designing a double stranded RNA effector molecule to selectively target said region of the non-coding strand replication intermediate, said rRNA effector molecule comprising an effector strand having a sequence of at least 19 contiguous nucleotides of an inverse complement of said region of the non-coding strand replication intermediate; and c) producing said dsRNA effector molecule or an expression construct that expresses said molecule.
