The invention relates to formulations and a method without chaotropic components, for the isolation of nucleic acids with binding to a solid phase, in particular, of DNA from complex starting materials containing a lysis/binding buffer system with at least one antichaotropic salt component, wherein the concentration of the antichaotropic salt component is between 0.001 mM and 0.1 M, preferably 0.1 mM, a solid phase and a known washing and eluting buffer. The lysis/binding buffer system can be present in the form of an aqueous solution or as a solid formulation in ready-to-use reaction vessels. All support materials which have application for isolation by means of chaotropic reagents can function as solid phases, preferably glass fibre matting, glass membranes, silicon supports, ceramics, zeolites, or materials with negatively-functionalised surfaces or chemically modified surfaces which can be made to carry a negative charge potential. The invention further relates to a method for isolation of nucleic acids, in particular of DNA, from complex starting materials using the inventive formulations, characterised by the lysis of the starting material, binding of the nucleic acid to a support material, washing the nucleic acids bonded to the support and elution of the nucleic acids.
