Provided is an optimized flagellin protein and a preparation method and use thereof. The optimized flagellin protein is a protein with deletion in the hypervariable region, which is the region of amino acids 180 to 400 of a flagellin protein, and the said protein includes FliCΔ190-278, FliCΔ220-320 and FliCΔ180-400. The optimized flagellin protein retains an adjuvant activity and has a reduced antigenicity and immunogenicity.
