Method of using a cereal protein which is not a storage protein as fusion carrier to highly express a small peptide in endosperm cells of the host, comprising the steps of: (i) providing the endosperm-specific promoter and the sequence signal peptide of SEQ ID NO: 1; (Ii) providing the gene of the protein of cereal non-storage protein as one fusion carrier and an target gene encoding the small peptide; (Iii) constructing an expression vector containing the leader DNA sequence and promoter, the gene of the fusion carrier and the target gene; and (iv) expressing the expression vector in a cell host endosperm, wherein the small peptide has fewer than 100 amino acids and wherein the cereal protein which is not a storage protein is selected from the group consisting of at the C-terminal binding protein endoplasmic reticulum (Bip) encoded by the nucleotide sequence of SEQ ID NO: 3, and the C-terminal of protein disulfide isomerase (PDI) encoded by the nucleotide sequence of SEQ ID NO: 4.
