PURPOSE: A biochip, a manufacturing method thereof and an object substance detecting method using the same are provided to simplify the manufacturing process and reduce the manufacturing costs by rapidly and easily forming the working electrode and reference electrode of a bio-chip through a lift-off process.;CONSTITUTION: A biochip(100) comprises a first substrate and a second substrate. In the first substrate, a working electrode(120) in which the aptamer coupling with a reference electrode(130) is able to be fixed is formed. The second substrate faces with the first substrate. A microfluidic channel(150) forming a flow passage on the working electrode and reference electrode is formed in the second substrate.
