PURPOSE: A method for preparing chitosan microsphere is provided to produce chitosan microsphere having improved elasticity and flexibility. CONSTITUTION: A method or preparing chitosan microsphere comprises: a step of making chitosan solution; a step of dropping chitosan solution to upper layer of organic solvent and lower layer of alkali solution or cationic solution; and a step of cross-linking the microsphere. The organic solvent which is not soluble with water is petroleum ether or ethel ether. Dropping is performed using nitrogen gas. The cross-linking is performed by reacting with genipin.
