A method for producing a biologically active polypeptide having a recognition site N-terminalHis-Gly with insulinotropic activity, the method comprising the steps of: a) transforming a genetically modified host cell having gene deletion proteasaSTE13 with a polynucleotide vector encoding the polypeptide; b) culturing the transformed host cell to produce the polypeptide; c) alpha-amidating the polypeptide to produce biologically active polypeptide.
