The present invention relates to the screening of molecules with anti-prion activity. It relates more particularly to screening kits for molecules with anti-prion activity, characterized in that they comprise in combination a yeast of phenotype [PSI +], an antibiogram and a prion curative agent at sub-effective doses, said yeast exhibiting the adel-14 allele of the ADE1 gene as well as an inactivated ERG6 gene, the screening methods, and a family of molecules with anti-prion activity demonstrated using the screen according to the invention. Application of anti-prion agents to manufacture medicaments, in particular for the treatment of neurodegenerative diseases involving protein aggregates.
