PURPOSE: A forced expression vector of foreign gene is provided to prepare a metagenome, genome or cDNA library, to screen a useful gene from the library, and to search a gene using a transcription/translation system in vitro.;CONSTITUTION: A gene forced expression cassette contains a promoter, lactose operator(lacO), and three-frame simultaneous translation(TFST) construct. The TFST construct comprises a ribosome binding site(RBS), RBS, initiation codon(ATG), RBS, initiation codon(ATG), and initiation codon(ATG) which are operatively linked in order. The TFST construct has a base of sequence number 14. The promoter is T7 promoter.
