PURPOSE: A development of technology for selecting a genome related to backfat in Hanwoo(Korean beef) is provided to early diagnose and select high quality Hanwoo using a DNA genotype marker. CONSTITUTION: A method for diagnosing meat traits of Hanwoo using a molecular marker related to the backfat thickness of Hanwoo comprises PCR-RFLP(Restriction Fragment Length Polymorphism) analysis comprising a step of treating an SPP1(secreted phosphoprotein 1) gene amplification product with an Nla III restriction enzyme. An individual DNA marker of Hanwoo by the substitution at a 92th base(C to T, T to C) of the SPP1 gene is detected through the method. Hanwoo with genetically thin backfat is determined by analyzing SNP genotype(CC, CT, TT) through the method. SPP1 gene is amplified using a forward primer of sequence number 2(5'-AGTGAGGAGATGCATGACGC-3') and a reverse primer in sequence number 3(5'-CATATTGTCTCCCACCCTGCT-3').
